{"id":214558,"date":"2024-06-06T07:15:48","date_gmt":"2024-06-06T11:15:48","guid":{"rendered":"https:\/\/today.uconn.edu\/?p=214558"},"modified":"2024-06-04T10:56:31","modified_gmt":"2024-06-04T14:56:31","slug":"uconn-professor-patents-novel-food-allergy-treatment","status":"publish","type":"post","link":"https:\/\/today.uconn.edu\/2024\/06\/uconn-professor-patents-novel-food-allergy-treatment\/","title":{"rendered":"UConn Professor Patents Novel Food Allergy Treatment"},"content":{"rendered":"<p>Clinton Mathias, associate professor of <a href=\"https:\/\/nusc.uconn.edu\">nutritional sciences<\/a> in the College of Agriculture, Health and Natural Resources (<a href=\"https:\/\/cahnr.uconn.edu\">CAHNR<\/a>), has discovered a potential new treatment for allergic reactions.<\/p>\n<p><img decoding=\"async\" class=\"alignleft wp-image-209097 img-responsive lazyload\" data-src=\"https:\/\/today.uconn.edu\/wp-content\/uploads\/2024\/01\/cahnr_10th_blue-green-300x139.png\" alt=\"CAHNR 10th Anniversary of Health badge\" width=\"180\" height=\"84\" src=\"data:image\/svg+xml;base64,PHN2ZyB3aWR0aD0iMSIgaGVpZ2h0PSIxIiB4bWxucz0iaHR0cDovL3d3dy53My5vcmcvMjAwMC9zdmciPjwvc3ZnPg==\" style=\"--smush-placeholder-width: 180px; --smush-placeholder-aspect-ratio: 180\/84;\" \/>Daniel Kennedy, Mathias\u2019 colleague at <a href=\"https:\/\/wne.edu\/\" target=\"_blank\" rel=\"noopener\">Western New England University<\/a> where he worked before joining the UConn faculty in 2022, had been studying thiol isomerases in a mouse cancer model. At the same time, Mathias had been working with curcumin, the active ingredient in the curry spice turmeric and investigating its effects in mouse models of food allergies. These studies showed that curcumin had potent inhibitory effects on the development of food allergy. When mice were exposed to curcumin during an allergic reaction, they did not develop allergic symptoms.<\/p>\n<p>The team published their findings in <a href=\"https:\/\/www.frontiersin.org\/journals\/immunology\/articles\/10.3389\/fimmu.2020.606837\/full\" target=\"_blank\" rel=\"noopener\">Frontiers in Immunology<\/a> in 2020.<\/p>\n<p>These protective effects were exerted by directly targeting a major immune culprit during food allergy reactions, the mast cell, and suppressing its activity.<\/p>\n<p>Thiol isomerases play critical roles in the functioning of cells and are important for their activity and survival. \u00a0They play key roles in the folding of cellular proteins and are known to promote the activity of platelets and control blood clotting reactions.<\/p>\n<p>Together, Mathias and Kennedy investigated whether curcumin may also block the function of thiol isomerases.<\/p>\n<p>\u201cWe wondered whether curcumin might also potentially inhibit this particular enzyme,\u201d Mathias says. \u201cAnd we found that it did. It inhibited the enzyme\u2019s catalytic activity. Furthermore, we found that mast cells can also express cell surface thiol isomerases, something that was previously shown only on a few cell types such as endothelial cells and hepatocytes.\u201d<\/p>\n<p>Immune cells such as mast cells produce many pro-allergic proteins during allergic reactions. Thiol isomerases may play a role in the folding and activity of these proteins. \u201cMany of these are extremely important to drive the allergic response,\u201d Mathias says. \u201cSo, if you block that, you\u2019re blocking the allergic response.\u201d<\/p>\n<p>Mathias tested the efficacy of PACMA-31,\u00a0 a protein disulfide isomerase (PDI) inhibitor in mice. Mathias treated the mice with the inhibitor every day for the extent of their exposure to the allergen, a protein that is commonly present in egg whites. The use of this protein, ovalbumin, is widely accepted as a research model for food allergies.<\/p>\n<p>\u201cIt\u2019s a pretty good indicator that this would also work in other types of allergies, at least in mice,\u201d Mathias says. \u201cIn humans, it remains to be tested.\u201d<\/p>\n<p>The inhibitor suppressed mast cell activation in the mice dramatically. The mice did not experience food allergy symptoms that were observed in the control group that did not receive the inhibitor.<\/p>\n<p>The patent for this technology was granted earlier this year and Quercis Pharmaceuticals has obtained a license for this technology. Western New England University is the assignee for this patent.<\/p>\n<p>The researchers still need to conduct additional studies to determine if the inhibitor just blocks allergic symptoms or if it can also prevent the development of sensitivity to the allergen.<\/p>\n<p>\u201cOur hypothesis is that it would,\u201d Mathias says. \u201cBecause, when we treat cells with the PDI inhibitors before we\u2019ve even activated them with the allergen, we find reduced activation. So, I think it\u2019s possible that it actually prevents the development of food allergy and not just treats it.\u201d<\/p>\n<p>Other PDI inhibitors are already being tested in clinical trials as cancer drugs and a handful of other applications.
